چکیده

Murine intestinal intraepithelial lymphocytes (IEL) from unprimed thymus-bearing and athymic nude mice were characterized according to the expression of murine lymphocyte antigenic markers and cytotoxic activity. The majority of IEL from thymus-bearing mice were Lyt-2+, L3T4- lymphocytes, over half of which did not express Thy-1 surface antigens. Nude IEL and spleen cells were void of Thy-1+ cells; however, Lyt-2 antigens were expressed on a significant proportion of IEL, but not splenic lymphocytes. Overall, 40-70% of IEL from either thymus-bearing or athymic mice expressed the cytotoxic activation antigen, CT-1, and the J11d lymphocyte marker, both of which were associated with a population of Thy-1-, Lyt-2+ cytotoxic IEL. These data are taken to mean that the intestinal epithelium is a site uniquely enriched for activated cytotoxic cells, a significant proportion of which originate as non-thymus-derived lymphocytes with acquired lytic activity.

Characteristics of natural killer cells in the murine intestinal epithelium and lamina propria

Highly purified populations of lymphocytes were obtained from the murine intestinal mucosa using EDTA-collagenase isolation procedures in combination with discontinuous density centrifugation. Intraepithelial lymphocytes (IEL) were separated from lamina propria lymphocytes (LPL) and, within these two populations, fractions enriched or depleted in gut granular lymphocytes (gGL) were obtained. Us...

Blocking effect of lyt-2 antibodies on T cell functions

Monoclonal anti-Lyt-2 antibodies blocked effector function of cytotoxic thymus-derived (T) cells in the absence of added complement. Cytolysis of both allogeneic cells and syngeneic lymphoma or sarcoma target cells was inhibited at the level of the effector lymphocytes. Anti-Lyt-1 and anti-Thy-1 antibodies did not block killer cells. Proliferation of T cells in mixed lymphocyte culture was also...

Characterization of interleukin 2-dependent cytotoxic T-cell clones: specificity, cell surface phenotype, and susceptibility to blocking by Lyt antisera.

Cytotoxic T-cells were derived from the peritoneal cavity of a C57BL/6 mouse immunized with BALB/c sarcoma Meth A and from the spleens of BALB/c x C57BL/6 F1 (hereafter called CB6F1) mice immunized with BALB/c leukemia RL male 1. The cells were cultured in interleukin 2 and cloned by limiting dilution, and their specificity was determined by direct tests and competitive inhibition assays. C57BL...

Cell Ontogeny : Organ Location of Maturing

Cytotoxic depletion studies with anti-Lyt-l , anti-Lyt-2, and anti-Lyt-3 antisera demonstrated that functional mouse T cell subpopulations differ in their expression of these surface antigens. Helper T cells were sensitive to depletion only with antiLyt-1, whereas suppressor and cytotoxic T cells were depleted only with anti-Lyt-2 or anti-Lyt-3 (1-3). Thus, helper T cells were classified as Lyt...

Qualitative and quantitative heterogeneity in Pgp-1 expression among murine thymocytes.

A proportion of Pgp-1+ cells in the thymus have been shown to have progenitor activity. In adult AKR/Cum mice the total Pgp-1+ population in the thymus differs from that of the bulk of thymocytes and is antigenically heterogeneous when examined by flow cytometry. Pgp-1+ thymocytes are enriched for several minor cell populations compared to total thymocytes: B2A2-, interleukin-2-receptor+ (IL-2R...
